Streptomyces bio-products against apple and pear diseases in organic orchards

Tinatin DOOLOTKELDİEVA

Erwinia amylovora as a fire blight pathogen and Venturia inaequalis as a scab pathogen were isolated from the blossoms, exudates, infected fruits, leaves and bent branches of diseased apple, pear and hawthorn trees, selected in the Chy, Osh and Jalal Abad regions. Biochemical and pathogenicity tests, alongside PCR analyses were conducted to identify the local isolates of Erwinia amylovora and Venturia inaequalis. The alternative antagonistic microorganisms which combat bacterium E. amylovora and fungus Venturia inaequalis were tested within in vitro and in vivo conditions. The results revealed ...Более

Occurrence, identification and phylogenetic analyses of cereal cyst nematodes in Kyrgyzstan

Şenol YILDIZ | Tair Esenalı Uulu

The study investigated the status of cereal cyst nematodes (CCN) in the main wheat-growing areas of Kyrgyzstan in 2020. Soil samples were taken from 69 different wheat fields located in Chuy and Issyk-Kul provinces. CCN were found in thirty-one out of the sixty-nine locations surveyed. The highest occurrence of CCN was in the Tyup location in Issyk-Kul province with 81 cysts (250 cm3 soil)-1. The CCN populations were identified by both morphological and molecular analyses. According to the results, all populations were identified as Heterodera filipjevi. No variations in rDNA-ITS sequencing da ...Более

Nematode-citrus plant interactions: host preference, damage rate and molecular characterization of Citrus root nematode Tylenchulus semipenetrans

Şenol YILDIZ

center dot Citrus plants are host to several plant parasitic nematodes (PPNs), which are microscopic organisms. Among PPNs, the citrus root nematode, T. semipenetrans (Cobb 1913) (Tylenchida: Tylenchulidae), causes significant damage to citrus plantations worldwide. Understanding citrus nematode populations, precise identification, host preference among citrus species, and damage threshold are crucial to control T. semipenetrans. The minutiae of citrus plant-nematode interactions, nematode density and molecular nematode identification are not well understood. In this study, nematode species an ...Более

Species Discrimination within the Metarhizium PARB Clade: Ribosomal Intergenic Spacer (rIGS)-Based Diagnostic PCR and Single Marker Taxonomy

Saykal BOBUŞOVA | Tinatin DOOLOTKELDİEVA

Background: The entomopathogenic fungus Metarhizium anisopliae sensu lato forms a species complex, comprising a tight cluster made up of four species, namely M. anisopliae sensu stricto, M. pinghaense, M. robertsii and M. brunneum. Unambiguous species delineation within this “PARB clade” that enables both the taxonomic assignment of new isolates and the identification of potentially new species is highly solicited. (2) Methods: Species-discriminating primer pairs targeting the ribosomal intergenic spacer (rIGS) sequence were designed and a diagnostic PCR protocol established. A partial rIGS se ...Более

Identification of Microbial Populations Present in Agricultural and Nonarable Soils in the Talas Valley, Northern Kyrgyzstan, in Autumn

Tinatin DOOLOTKELDİEVA | Saykal BOBUŞOVA | Mahabat KONURBAYEVA

Soil bacterial and fungal communities were investigated in relation to soil type and farm management practices after vegetation harvesting in autumn. Soils from fields cultivated with Phaseolus vulgaris (bean) and Pyrus comminus (pear) and nonarable, natural areas were studied. Microbial diversity was analysed using cultivation-dependent methods (isolation of pure cultures) and cultivation-independent methods (direct extraction of DNA from soil, followed by PCR amplification of the 16S rRNA and 18S rRNA genes). The use of cultivation-dependent methods revealed that there were no differences in ...Более

First record of tomato leafminer Tuta absoluta Meyrick (Lepidoptera: Gelechiidae) in Kyrgyzstan

Tair Esenalı Uulu

This short note reports the first record of the tomato leaf miner Tuta absoluta Meyrick (Lepidoptera: Gelechiidae) in Kyrgyzstan. This pest was found in February 2017 in greenhouses producing tomatoes, located 15 km from Bishkek. Identification of the pest was carried out in the Plant Protection Department, Çukurova University, Turkey based on the morphology of adults and larvae and the male genitalia. According to initial surveys this pest could be a major threat to tomato production in Kyrgyzstan.

Effects of some plant extracts on root-knot nematodes in vitro and in vivo conditions

İlker KEPENEKÇİ

One of the major pests of the vegetables, root knot nematodes (Meloidogyne spp.) (Tylenchina: Meloidogynidae) (RKNs) can cause economic losses by forming knots on the host plant roots. RKNs are more prevalent in the greenhouse vegetable growing areas of the coastal regions. In this study, the effects of plant extracts from five different plants; Capsicum frutescens, Hyoscyamus niger (Solanaceae), Melia azedarach (Meliaceae), Xanthium strumarium and Achillea wilhelmsii (Asteraceae) were evaluated against RKNs. In the first studies; the effects of plant extracts (0.5, 1.0, 1.5, 3.0, 6.0 and 12.0 ...Более

Biological Control Potential of Turkish Entomopathogenic Nematodes Against Holotrichapion pullum (Gyllenhal) (Coleoptera, Apionidae)

İlker KEPENEKÇİ

Potential of three native Turkish isolates of entomopathogenic nematodes (EPNs) (Steinernema feltiae, S. carpocapsae and Heterorhabditis bacteriophora) was evaluated against Holotrichapion pullum (Gyllenhal) (Coleoptera, Apionidae) under laboratory conditions. Suspensions of EPNs were applied at three concentrations (500, 1000 and 2000 IJs ml-1) and at two temperatures (15 and 20°C). After one week, mortality rate of H. pullum adults was assessed. Mortality rate was higher in the nematode treatments than in the control. All nematodes showed more effect depending on temperature increase. At 20° ...Более

Biological Control Agents for Crop Protection and Sustainability of Agro-ecosystems in Kyrgyztan

Tinatin DOOLOTKELDİEVA

The main agricultural activities in the Kyrgyz Republic are include the production of cereal grains, vegetables, potatoes and commercial crops such as cotton, tobacco, sugar beets and livestock. The sustainability of the agriculture sector was severely affected by the large scale mono-cropping approach of the Soviet era. Intensive agriculture has caused environmental pollution due to the excessive use of mineral fertilizers and pesticides. To combat this menace we have isolated and introduced non-chemical crop protection agents, the antifungal micro-organisms Streptomyces spp., producing antib ...Более

Microbial communities of vegetable seeds and biocontrol microbes for seed treatment

Tinatin DOOLOTKELDİEVA | Saykal BOBUŞOVA

Thirty-one species of vegetable seeds from Kyrgyzstan's State Register were analysed for mycological and bacterial composition using various methods, from simple visual to molecular. The modified roll-towel method simultaneously assesses the degree of contamination and seed germination. Screening in agar medium allows analysis of species on the outer and inner surfaces of vegetable seeds that are not detected using the roll-towel method. As a result of the phytopathological examination, 15 fungal and three bacterial species were found. Direct DNA extraction and amplification of the universal 1 ...Более
