Characterization of a 2,2-dichloropropionic acid (2,2-dcp) degrading alkalotorelant bacillus megaterium strain bhs1 isolated from blue lake in Turkey

Yılmaz KAYA

An acid, 2,2-dichloropropionic acid (2,2-DCP) is an active ingredient in herbicide (Dalapon®). Using 2,2-DCP as a model substrate, an alkalotolerant bacterium was successfully isolated from the Blue Lake, Turkey. This bacterium is a potential bioremediation agent of recalcitrant xenobiotic halogenated compounds. This study aimed to prove the efficacy of the alkalotolerance Bacillus megaterium BHS1 in degrading 2,2-DCP as the sole source of carbon. Biolog GEN III system and 16S rRNA analysis were used for the identification of the bacterium. It was discovered that the strain BHS1 is Bacillus m ...Daha fazlası

Comparative modeling and enzymatic affinity of novel haloacid dehalogenase from Bacillus megaterium strain BHS1 isolated from alkaline Blue Lake in Turkey

Yılmaz KAYA

This study presents the initial structural model of L-haloacid dehalogenase (DehLBHS1) from Bacillus megaterium BHS1, an alkalotolerant bacterium known for its ability to degrade halogenated environmental pollutants. The model provides insights into the structural features of DehLBHS1 and expands our understanding of the enzymatic mechanisms involved in the degradation of these hazardous pollutants. Key amino acid residues (Arg40, Phe59, Asn118, Asn176, and Trp178) in DehLBHS1 were identified to play critical roles in catalysis and molecular recognition of haloalkanoic acid, essential for effi ...Daha fazlası

Production of secondary metabolites using tissue culture-based biotechnological applications

Yılmaz KAYA

Plants are the sources of many bioactive secondary metabolites which are present in plant organs including leaves, stems, roots, and flowers. Although they provide advantages to the plants in many cases, they are not necessary for metabolisms related to growth, development, and reproduction. They are specific to plant species and are precursor substances, which can be modified for generations of various compounds in different plant species. Secondary metabolites are used in many industries, including dye, food processing and cosmetic industries, and in agricultural control as well as being use ...Daha fazlası

The effect of leaf blight disease of rice caused by Pantoea ananatis on Nikita, Osr30 and RIRE1 retrotransposons’ movements

Yılmaz KAYA

Changing environmental conditions can cause diseases in plants, leading to novel interactions between plants and stress factors. In this respect, the relationships between pathogens and plants regarding epigenetics have still been investigated. Our aim is to examine the effect of leaf blight bacterial causal agent, Pantoea ananatis isolate PaTo34a1, on barley- and rice-specific retrotransposons’ (Nikita, Osr30 and RIRE1, respectively) movements in different rice cultivars (Oryza sativa L. cv. Tosya, Cameo, Arize, Ronaldo, Osmancik-97, Kale, Baldo, Bafra, Hakkari and Vasco) collected from diffe ...Daha fazlası

A 2,2-dichloropropionic acid-degrading novel Pseudomonas fluorescence strain fatsa001: isolation, identification, and characterization

Gülbübü KURMANBEKOVA | Yılmaz KAYA

There are mounting concerns over the high concentrations of non-biogenic, toxic halogenated organic compounds being liberated into the ecosystem. Therefore, this study’s isolation of a novel bacterium from a contaminated stream in Fatsa, Ordu, Turkey, adept in degrading 2,2-dichloropropionic (of 2,2-DCP) is a welcome endeavor. The ability of the bacterial isolate to utilize 2,2-DCP as the sole carbon and energy source was discovered when the bacterium was observed to grow well on liquid minimal media containing 20 mM of 2,2-DCP, showing a doubling time of 14.2 h. The following genetic and bioc ...Daha fazlası

Agrobacterium- mediated transformation of Turkish upland rice (Oryza sativa L.) for Dalapon herbicide tolerance

Yılmaz KAYA | İbrahim İlker ÖZYİĞİT | Ali ASLAN

Agrobacterium-mediated transformation of upland rice is established in few numbers of cultivars due to the high cultivar-specificity of regeneration from transformed explants. Further, dehalogenase E (dehE) gene had been characterized in Pseudomonas putida and it produces an enzyme that degrades dalapon. This study aimed to transform Turkish upland rice with the dehE herbicide resistant gene and addresses the challenges of transgenic rice recovery by identifying explant and transformation method. Constructed vector pCAMdehE carrying dehE gene was transferred into the rice shoot apex by Agrobac ...Daha fazlası

Genomic analysis of a functional haloacid-degrading gene of Bacillus megaterium strain BHS1 isolated from Blue Lake (Mavi Golu, Turkey)

Yılmaz KAYA

Purpose: Bacillus megaterium strain BHS1, isolated from an alkaline water sample taken from Mavi Golu (Blue Lake, Turkey), can grow on minimal medium containing 2,2-dichloropropionic acid. We characterized this bacterium at the genomic level. Methods: The HiSeq platform was used to carry out genome sequencing, de novo assembly, and scaffolding with strain BHS1. Next, genome data were analyzed to demarcate DNA regions containing protein-coding genes and determine the function of certain BHS1 genes. Finally, results from a colorimetric chloride ion-release assay demonstrated that strain BHS1 pr ...Daha fazlası

Identification of Dalapon degrading bacterial strain, Psychrobacter sp. TaeBurcu001 isolated from Antarctica

Yılmaz KAYA

Extreme microorganism has gained interest because of their special contribution to the environmental and industrial biotechnology, particularly psychrophilic microorganisms which can live and thrive at lower temperatures. They produce enzymes with great potential to adapt to extreme conditions. In this study, a novel psychrophilic bacterial strain designated as TaeBurcu001 grew (optimally at 5.C) on a chemically defined medium containing dalapon (refer to as 2,2-dichloropropionic acid, 2,2-DCP) as the sole carbon source. TaeBurcu001 was isolated from Galindez Island, Antarctica. Phylogenetic a ...Daha fazlası

Antibacterial activity of Calendula officinalis and Echinacea purpurea extracts against the causal agent of tomatoes' bacterial canker: Clavibacter michiganensis subsp

Yılmaz KAYA

We aimed to investigate the effects of Calendula officinalis and Echinacea purpurea extracts in terms of growth parameters, antibacterial activity and phenolic profile in tomato infected by Clavibacter michiganensis subsp. michiganensis (CmmT7). A significant difference was observed in E. purpura extract, indicating the highest effects on plant height (27.25 cm), fresh plant weight (28.45 cm), root length (24.42 cm), and root weight (6.74 g) (p

Further Analysis of Burkholderia pseudomallei MF2 and Identification of Putative Dehalogenase Gene by PCR

Yılmaz KAYA

Halogenated organic compounds are extensively and widely used as pesticides, herbicides, and antibiotics that contribute to the pollution. This research was aimed to further analyze and characterize a bacterium that has the ability to utilize 2,2-dichloropropionic acid (2,2-DCP) as a model to study dehalogenase enzyme production. Microscopic observation, biochemical tests and PCR technique were carried out in order to characterize the isolated bacterium. Strain MF2 showed its ability to grow on 10 mM 2,2-DCP liquid minimal medium with doubling time of 13 h with maximum chloride ion released of ...Daha fazlası
